Long Time-lapse Experiments

A common problem with long time-lapse experiments has been that objects drift out of Z focus due to physical changes in the microscope stand and stage as a result of environmental conditions. To address this, softWoRx has the ability to allow periodic adjustments to be done during the time-lapse wait interval. To accomplish this, you can specify an UltimateFocus Action to be triggered for all time points, and further to use the When control to tell the system to perform the focus maintenance periodically During the time-lapse wait. It will be necessary to toggle the Maintain Focus with UltimateFocus button Off in the Time-lapse tab as the Action will be sufficient to maintain the focus. This should provide maximum flexibility to minimize Z drift focus artifacts in the experiment.
