Use Line Profile to view a plot of intensity values for pixels in a row or column of the Image window. You can view a profile of a line of pixels or a band of pixels and save profile data to a text file.
Open Line Profile by choosing Tools | Line Profile from the Image window.

Enter the window number of the Image window in which you want to view the line profile. If you are working with multiple Image windows and want to change the window number, you can type the new window number into the Window field or right-click the window number in the Image window and drag it to the Window field. |

AutoScale
|
Scale the profile to fit in the Image window. When AutoScale is off, values in the AutoScale field (0 and 65535 in the figure above) are used as minimum and maximum values for the plot. You can change these values by typing values in the field. |

Display the minimum and maximum values of the line profile. |

Direction |
Choose the axis for the profile. Choose Horizontal to display the profile along a horizontal axis in the Image window or choose Vertical to display the profile along a vertical axis. |

Band |
Specify that the profile shows the average intensity of a band of pixels in the image by typing a value other than zero. For example, you can choose a band to be the average diameter of spots in a single row. This allows you to view a profile of average intensities of wavelengths for spots in the row. If the band value is zero, the profile shows intensity values of single pixels in a row or column. You can measure the diameter of a spot by using Measure Distances or Point Values. |

XY

Change the row or column of pixels that are used to generate the profile.
A straight line with an 'X' on it in the Image window indicates the row or column of pixels in the Image window from which intensity values are obtained to generate the profile. The line is horizontal when Horizontal is selected in the Direction option list, and vertical when Vertical is selected. The values in XY show the position of the straight line in pixels, where 0,0 is the left and bottom of the image. Only the first number (X) changes when Direction is Vertical. The second number (Y) changes when Direction is Horizontal. You can change the position of the straight line by using one of the following methods:

AutoSave |
Turn on AutoSave to save line profiles as they are displayed in the Image window. For example, you can save the line profiles of several rows of bands of pixels that show the average intensities of spots in the row by turning on AutoSave and displaying a line profile for each row of interest. You can create a text file of the data in the saved profiles by clicking Write to File. Note If you want to save line profiles, but do not want to save every profile as it is displayed, you can save the profile displayed in the Image window by clicking Save. |

Profiles Saved |
View the number of profiles currently saved in memory. This is the number of profiles that are saved in a text file if you save the profiles to a file. |

Save |
Save the profile displayed in the Image window (If AutoSave is off). You can create a text file of the data. Profiles are saved by clicking Write to File. |

Clear |
Clear all saved profiles from memory. |

Write to File
|
Open the Profile Output Options window and save the data to a text file. If the number in Profiles Saved is greater than zero, profiles are saved in memory. |

Done |
Close the Line Profile window. |
