Time-lapse Options

Use the Time-lapse tab fields to specify the number of time points, the time periods, and the total time for a time-lapse experiment. (The Time-lapse tab is under the Design tab in the Design/Run Experiment window.) Like other macros, Time-lapse experiments should be designed after you have manually acquired images of your specimen and adjusted exposure and other parameters until you are satisfied with the images.

Note:  You'll need to specify only two of the three fields in this section. After you specify these fields, softWoRx calculates a value for the third field. In most cases, you'll enter a value in the Time-lapse field and then enter a value in either the Time Points or Total Time field.

 

 

Use

Time-lapse (check box)

Activate the check box to enable a time-lapse experiment.

Time-lapse

Specify the hours, minutes, and seconds for each time period.

Total Time

Displays the total time of the experiment, which can also be calculated as follows:

Total Time = (Number of time points - 1) x Time-lapse

Time Points

Specify the number of time samples to collect in a time-lapse experiment.

Enable Cell Tracking

Move the stage laterally to follow cells as they move during a time-lapse experiment. With the Enable Cell Tracking option selected, DeltaVision automatically keeps cells in the field of view.

Moves the stage laterally to follow cells as they move during a time-lapse experiment. With this check box activated, DeltaVision automatically keeps cells in the field of view.

Cell Tracking Options

Opens the Cell Tracking Options window that allows you to specify the parameters for cell tracking.

Maintain Focus with UltimateFocus (check box)

Specifies focus should be maintained during the time-lapse experiment. If this experiment does not involve point visiting, a calibration (characterizing the UltimateFocus response at the current location) is performed when the experiment is started.

Notes:  #1 The initial focus is essential to the success of focus maintenance.  #2 The actual focus maintenance process happens just before each time-lapse event.

UltimateFocus Options

Opens the UltimateFocus Options window, in which you can indicate a specific Move Threshold and the Maximum (number of) Iterations to move within the threshold before re-running UltimateFocus.

•  In the Move Threshold field, enter the desired maximum measured focus error (in mm) before a corrective action should be taken.

•  In the Maximum Iterations field, enter the maximum measure/move sequences for this action to reach the calibrated focus point.

Image-based Autofocus before imaging (check box)

Automatically focus the camera before acquiring each time point. (This option does not cause DeltaVision to AutoFocus between wavelengths or Z steps.) For point visiting experiments, the camera is automatically focused every time a point is visited.

Autofocus Options

Opens the Resolve3D Settings window to the Autofocus tab, in which you can manipulate the settings for the software-based Autofocus function.
