To acquire a reference image:

Use an alternate filter or the transmitted light to acquire a reference image that can be combined with other images. This option is useful for Differential Interference Contrast (DIC) analysis. It can also be useful for other types of reference images.

  1. Manually acquire an image.

  2. In the Design/Run Experiment window, click the Channels tab and select the channels for your experiment.

  3. Activate the Reference Image check box.

  4. In the Z Position list, choose whether to acquire the reference image at the top, middle, or bottom of the sample.

  5. Under the Reference Image check box, select which filters to use for the reference image.

  6. Select the type of light to use for the reference image (EX is the excitation light source (EX1 and EX2 for multiplex wavelength configurations), TRANS is the transmitted light, and LASER is for laser light sources).

  7. Click the Run Experiment tab and click the Start Scan Run_icon_sm.JPG button to start the experiment.

    DeltaVision saves the experiment data in two files. One file includes only the reference image data and the other file includes all of the other data acquired in the experiment. By default, the reference file is not displayed.

  1. Tip:  After you acquire the images, you can use the Image Fusion tool in softWoRx to combine the two files together.
