To apply a calibration table:
Remove the calibration slide.
Mount the sample and focus.
Acquire an image.

Note: Consistent illumination is important for quantitative image processing. Calibration should be repeated whenever the illumination optics are modified. That is, the optical path should be identical during generation of the calibration table and the acquisition of the image data files. The same lamp, filters, and lenses should be used. In addition, each wavelength that is examined should have a separate calibration table. |
